Background: Luteinizing Hormone(LH) is a protein hormone secreted by the pituitary gland and transported through the bloodstream to the reproductive system. In the female body, LH stimulates the growth of follicles in the ovary and the release of mature eggs, as well as the production of progesterone to support conception and maintain pregnancy. In the male body, LH stimulates the testes to produce testosterone to support reproductive function and sexual maturity.LH levels typically vary with the menstrual cycle and age. In the female body, LH levels differ at different stages of the menstrual cycle, such as reaching a peak during ovulation. Principle of the Assay: This kit was based on Competitive-ELISA detection method. The microtiter plate provided in this kit has been pre-coated with LH. During the reaction, LH in the sample or standard competes with a fixed amount of LH on the solid phase supporter for sites on the Biotinylated Detection Antibody specific to LH. Excess conjugate and unbound sample or standard are washed from the plate, and HRP-Streptavidin (SABC) is added to each microplate well and incubated. Then TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm. The concentration of LH in the samples is then determined by comparing the OD of the samples to the standard curve. The concentration of the target substance was inversely proportional to the OD450 value.
