Background: Immunoglobulin E (IgE) is a type of antibody (or immunoglobulin (Ig) "isotype") that has only been found in mammals. IgE is synthesised by plasma cells. Monomers of IgE consist of two heavy chains (epsilon chain) and two light chains, with the epsilon chain containing 4 Ig-like constant domains (Cepsilon1-Cepsilon4). IgE's main function is immunity to parasites such as helminths like Schistosoma mansoni, Trichinella spiralis, and Fasciola hepatica. IgE is utilized during immune defense against certain protozoan parasites such as Plasmodium falciparum.
Principle of the Assay: The Mouse IgE ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse IgE in Cell Culture Supernates, Serum, Plasma, Cell Lysates, Tissue Homogenates. This assay employs an antibody specific for Mouse IgE coated on a 96-well plate. Standards and samples are pipetted into the wells and IgE present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse IgE antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IgE bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
