Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-GPR54/KISS1R antigen affinity purified polyclonal antibody (#AAA19421) at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for GPR54/KISS1R at approximately 80 kDa. The expected band size for GPR54/KISS1R is at 80 kDa.)

Background: Carcinoembryonic antigen-related cell adhesion molecule 8 (CEACAM8) also known as CD66b (Cluster of Differentiation 66b), is a member of the carcinoembryonic antigen (CEA) gene family. CEACAM-8 (Carcinoembryonic Antigen-related Cell Adhesion Molecule 8), also known as CD66b, CD67 and NCA-95, is a 90 kDa member of the CEACAM subfamily of the CEA family of proteins. It is expressed by neutrophils and eosinophils, and serves as a binding partner for CEACAM-6 and Galectin-3. Mature human CEACAM-8 is a 287 amino acid GPI-linked glycoprotein. It contains one Vtype and two C2-type Ig-like domains. No definitive rodent CEACAM-8 has been reported.

Principle of the Assay: The Picokine™ Human CEACAM8 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Human CEACAM8 with a 96-well strip plate that is pre-coated with antibody specific for CEACAM8. The detection antibody is a biotinylated antibody specific for CEACAM8. The capture antibody is a monoclonal antibody from mouse and the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Human CEACAM8 with immunogen: Expression system for standard: NS0; Immunogen sequence: Q35-D320. The kit is analytically validated with ready-to-use reagents. To measure Human CEACAM8, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Human CEACAM8 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human CEACAM8 in the sample.
