In muscle physiology, plays a central role inthe balance between atrophy andhypertrophy. When recruited by MSTN,promotes atrophy response viaphosphorylated SMAD2/4. MSTN decreasecauses SMAD4 release and subsequentrecruitment by the BMP pathway to promotehypertrophy via phosphorylated SMAD1/5/8.Acts synergistically with SMAD1 and YY1 inbone morphogenetic protein (BMP)-mediatedcardiac-specific gene expression. Binds toSMAD binding elements (SBEs)(5'-GTCT/AGAC-3') within BMP responseelement (BMPRE) of cardiac activatingregions (By similarity). Common SMAD(co-SMAD) is the coactivator and mediator ofsignal transduction by TGF-beta (transforminggrowth factor).
