Background: 27-Hydroxycholesterol (27-HC) is an endogenous oxysterol with multiple biological functions, including activity as a selective estrogen receptor modulator (SERM) (a mixed, tissue-specific agonist-antagonist of the estrogen receptor (ER)) and as an agonist of the liver X receptor (LXR). It is a metabolite of cholesterol that is produced by the enzyme CYP27A1. A link between high cholesterol and breast cancer has been identified, and it has been proposed that this is due to 27-HC production by CYP27A1.
Principle of the Assay: Human 27-hydroxycholesterol (27-HC) ELISA Kit employs a two-site sandwich ELISA to quantitate 27-HC in samples. An antibody specific for 27-HC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any 27-HC present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human 27-HC detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of 27-HC bound in the initial step. The color development is stopped and the intensity of the color is measured.
