Background: Glucagon is an insulin-reactive hormone that is mainly produced by islet alpha cells. Its main role is to raise blood glucose level and release glycogen stored in the liver into the blood by promoting the liver glycogenolysis. This is important for maintaining constant blood glucose level, as hypoglycemia can lead to dangerous symptoms such as dizziness, rapid heartbeat, fatigue and even coma etc. When the body needs to replenish energy, insulin level decreases, meanwhile glucagon level secreted by islet alpha cells increases. Thus, glycogen in the liver is gradually decomposed to glucose, which is then released into the blood and supplied to parts of the body that need energy, such as muscle. In addition, glucagon can also have other effects on the metabolism of the body through other mechanisms, such as inhibiting gastrointestinal peristalsis and gastric acid secretion, and promoting the oxidative decomposition of fatty acids etc.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti GC antibody was pre-coated onto the 96-well plate. The biotin conjugated anti GC antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with GC conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of GC in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
