Principle of the Assay: Cu2+ can be reduced to Cu+ by protein under alkaline conditions. Cu+ can combine with BCA reagent and form purple complex, which has a maximum absorption peak at 562 nm. The OD value is proportional to the protein concentration. The protein concentration can be calculated indirectly according to the OD value at 562 nm.

What are protein assays?


Protein assays are quantitative techniques that measure total protein concentration in samples using colorimetric or fluorometric detection methods. BCA assays use copper ion reduction followed by bicinchoninic acid chelation for sensitive protein quantification.​


What is the BCA protein assay kit?


The BCA (Bicinchoninic Acid) protein assay kit is a colorimetric assay based on copper ion reduction by proteins in alkaline conditions, forming a purple complex detectable at 562 nm for accurate protein concentration measurement.​


What is the detection range and sensitivity of the protein assay?


BCA assay detection ranges from 20–2,000 µg/mL with standard protocols, or 125–2,000 µg/mL for microplate formats. Enhanced protocols shift the range to 5–250 µg/mL for more dilute samples with improved sensitivity.​


Does the kit work with detergents or reducing agents?


Standard BCA assays are compatible with most ionic and non-ionic detergents but incompatible with reducing agents like DTT. However, reducing agent-compatible versions are available with a compatibility reagent allowing up to 5 mM DTT.​


How long does the protein assay take to complete?


The rapid BCA assay completes in 5 minutes at room temperature, while standard protocols require 30 minutes at 37°C or 2 hours at room temperature, depending on the kit formulation and incubation conditions.​


What storage conditions are required for the reagents?


BCA assay reagents are typically stored at room temperature with light protection for 12 months. Reconstituted compatibility reagent, if used, remains stable for 8 hours at 4°C when protected from light.​
