Background: Bone morphogenetic protein 5 is a protein that in humans is encoded by the BMP5 gene.1,2,3 The protein encoded by this gene is member of the TGFbeta superfamily. Bone morphogenetic proteins are known for their ability to induce bone and cartilage development. BMP5 may play a role in certain cancers. Like other BMP's BMP5 is inhibited by chordin and noggin. It is expressed in the trabecular meshwork and optic nerve head and may have a role in the development and normal function. It is also expressed in the lung and liver. BMP-5 is mapped on Bovine chromosome 6.4 The standard used in this kit is recombinant BMP-5 with the monomer molecular mass of 18KDa.
Principle of the Assay: To measure Bovine BDNF, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Bovine BDNF in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Bovine BDNF in the sample.

The detection antibody is a biotinylated antibody specific for BDNF. The capture antibody is monoclonal antibody from mouse and the detection antibody is monoclonal antibody from mouse. The kit includes Bovine BDNF protein as standards.
