Principle of the Assay: The AAA Biotech' Human Carbonic Anhydrase 9 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Carbonic Anhydrase 9 in Cell Culture Supernatants, Serum, Plasma, Urine. This assay employs an antibody specific for Human Carbonic Anhydrase 9 coated on a 96-well plate. Standards and samples are pipetted into the wells and Carbonic Anhydrase 9 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Carbonic Anhydrase 9 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Carbonic Anhydrase 9 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Carbonic anhydrases (CAs) are a large family of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide. They participate in a variety of biological processes, including respiration, calcification, acid-base balance, bone resorption, and the formation of aqueous humor, cerebrospinal fluid, saliva, and gastric acid. This gene is mapped to 9p13.3. CA IX is a transmembrane protein and the only tumor-associated carbonic anhydrase isoenzyme known. It is expressed in all clear-cell renal cell carcinoma, but is not detected in normal kidney or most other normal tissues. It may be involved in cell proliferation and transformation.
