Background: Chitinase-3-like protein 1, also called CHI3L1 or YKL-40 is a secreted glycoprotein that is approximately 40kDa in size that in humans is encoded by the CHI3L1 gene. This gene is mapped to 1q32.1. Chitinases catalyze the hydrolysis of chitin, which is an abundant glycopolymer found in insect exoskeletons and fungal cell walls. The glycoside hydrolase 18 family of chitinases includes eight human family members. This gene encodes a glycoprotein member of the glycosyl hydrolase 18 family. The protein lacks chitinase activity and is secreted by activated macrophages, chondrocytes, neutrophils and synovial cells. The protein is thought to play a role in the process of inflammation and tissue remodeling.

Principle of the Assay: The Human YKL-40 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human YKL-40 in Cell Culture Supernatants, Serum, Plasma, Urine. This assay employs an antibody specific for Human YKL-40 coated on a 96-well plate. Standards and samples are pipetted into the wells and YKL-40 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human YKL-40 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of YKL-40 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
