Background: Coronaviruses are enveloped viruses with a positive-sense RNA genome and with a nucleocapsid of helical symmetry. Coronavirus nucleoproteins localize to the cytoplasm and the nucleolus, a subnuclear structure, in both virus-infected primary cells and in cells transfected with plasmids that express N protein. Coronavirus N protein is required for coronavirus RNA synthesis, and has RNA chaperone activity that may be involved in template switch. Nucleocapsid protein is a most abundant protein of coronavirus. During virion assembly, N protein binds to viral RNA and leads to formation of the helical nucleocapsid. Nucleocapsid protein is a highly immunogenic phosphoprotein also implicated in viral genome replication and in modulating cell signaling pathways. Because of the conservation of N protein sequence and its strong immunogenicity, the N protein of coronavirus is chosen as a test tool.
Principle of the Assay: The principle of this ELISA kit is based on the solid phase sandwich enzyme immunoassay technique. A monoclonal antibody specific for SARS-CoV-2 Omicron (B.1.1.529) variant Nucleocapsid has been pre-coated onto well plate strips. Standards and samples are added to the wells and SSARS-CoV-2 Omicron (B.1.1.529) variant Nucleocapsid present in the sample is bound by the immobilized antibody. After incubation the wells are washed and a horseradish peroxidase conjugated anti-SARS-CoV-2 Omicron (B.1.1.529) variant Nucleocapsid antibody is added, producing an antibody-antigen-antibody "sandwich complex". Following a wash to remove any unbound antibody a TMB substrate solution is loaded and color develops in proportion to the amount of SARS-CoV-2 Omicron (B.1.1.529) variant Nucleocapsid bound. The reaction is stopped by the addition of a stop solution and the intensity of the color can be measured at 450 nm (See schematics below).
