Directions per Liter
Dissolve 10.4g in 900ml of ddH2O, stirring until completely solubilized. Do NOT heat. Add 2g sodium bicarbonate with stirring. Adjust pH of the medium to 0.1-0.3 pH units below the desired pH. Add additional water to bring the solution to 1L. Filter-sterilize using a 0.22um membrane. Aliquot into sterile containers
Note: It may be necessary to lower the pH to 4.0 with 1N HCl to completely dissolve this product. After it has dissolved completely, the pH can be raised to 7.2 with 1N NaOH prior to the addition of sodium bicarbonate.

Preparation and Storage
Store powdered media at RT. Stable for 12 months after receipt. Opened bottles should be capped tightly and kept in a dark, low humidity environment. Prepared media should be kept at 4 degree C and used within a short period of time.

After dissolution, adjust pH to 7.2 using sodium bicarbonate or hydrochloric acid, filter-sterilize (0.22 μm), and store at 2-8°C. Aliquots can be frozen at -20°C for long-term storage.​


Are RPMI and DMEM the same?


No, RPMI 1640 and DMEM are distinct media formulations. RPMI 1640 contains lower glucose (2,000 mg/L) and higher amino acid concentrations, making it ideal for lymphoid cells. DMEM contains higher glucose (5,500 mg/L) and fewer amino acids, better suited for fibroblasts and epithelial cells.​
