Background: Human Defensin alpha 1 (DEFA1) is 94 amino acids (aa) in length and is cleaved into the following chains: HP 1-56 (aa 39-94), Neutrophil defensin 1 (aa 65-94) and Neutrophil defensin 2 (aa 66-94). Defensins are secreted peptides that kill microbes by permeabilizing their plasma membrane. DEFA1 has antimicrobial activity against Gram-negative and Gram-positive bacteria. ADP-ribosylation reduces cytotoxic and antibacterial activities, it is also known to enhance IL8 production.

Principle of the Assay: The Human Defensin alpha 1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Defensin alpha 1 in Cell Culture Supernates, Serum, Plasma, Cell Lysates, Tissue Homogenates. This assay employs an antibody specific for Human Defensin alpha 1 coated on a 96-well plate. Standards and samples are pipetted into the wells and Defensin alpha 1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Defensin alpha 1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Defensin alpha 1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
