Usage Notes
96 Well Clear Flat Bottom UV-Transparent Microplate is needed.
Do not mix or substitute reagents or materials from other kit lots or vendors.
The sample preparation must be done on ice to prevent the enzyme inactivation, and the enzyme activity should be determined on the same day. avoid repeated freeze-thaw.
Ensure all reagents and equipment are at the appropriate temperature before starting the assay.
The alpha-Glucosidase Activity Assay Kit (micro assay) provides a simple method for the detection of glycogen synthase activity in biological samples such as plant and animal tissues, cells and serum (plasma). In the assay, p-nitrophenol, which is present in the sample, has a maximum absorption peak at 400 nm, and alpha-GC activity is calculated by determining the rate of increase in absorbance value.

Principle of the Assay: alpha-Glucosidase (3.2.1.20, EC) widely exists in animals, plants, microorganisms and cultured cells. It catalyzes the hydrolysis of alpha-glucosidic bonds between aryl or hydrocarbyl groups and glycosyl groups to produce glucose, which is not only related to the relaxation or reinforcement of cell walls, but also closely related to cell recognition and the production of some signal molecules. alpha-GC decomposes p-nitrobenzene-alpha-D-glucopyranoside to generate p-nitrophenol, which has the maximum absorption peak at 400 nm. alpha-GC activity is calculated by measuring the rising rate of absorbance value.
