Biological Activity
Determine by its ability to chemoattract human blood monocytes. Chemotactic activity was observed at a concentration of 2.5 ug/ml with a peak response obatined at 250 ug/ml.
Reconstitution
Human Galectin-I should be reconstituted in water to a concentration of 0.1 mg/mI. This solution can be diluted in water or other buffer solutions or stored at -20 degree C.

Preparation and Storage
The lyophilized human Galectin-l, though stable at room temperature, is best stored desiccated below O degree C. Reconstituted human Galectin-I should be stored in working aliquots at -20 degree C . Avoid repeated freeze and thaw cycles!

Lectins, of either plant or animal ongm, are carbohydrate binding proteins that interact with glycoprotein and glycolipids on the surface of animal cells. Galectin-I (Gal-I) is one of 15 evolutionarily conserved 13-galactoside-binding proteins that display biologicallydiverse activities in pathogenesis of inflammation and cancer. Gal-I is variably expressed on immune cells and endothelial cells, though is commonly found and secreted at high levels in cancer cells. There is also compelling evidence in models of cancer and autoimmunity that recombinant Gal-I (rGal-l) can potentiate immunoregulatory function of T cells. Galectin-I is an animal lectin that has been shown to interact with CD3, CD4, and CD45. It induces apoptosis of activated T-cells and T-Ieukemia cell lines through homodimeric binding of Nacetyllactosamines on membrane glycoproteins (Gal-l ligands) and inhibits the protein phosphatase activity of CD45. Recombinant human Galectin-l is a 14.5 kDa protein containing 134 amino acid residues.
