Intended Uses: This F13B ELISA kit is intended for laboratory research use only. The stop solution changes the color from blue to yellow and the intensity of the color is measured at 450 nm using a spectrophotometer. In order to measure the concentration of F13B in the sample, this F13B ELISA Kit includes a set of calibration standards. The calibration standards are assayed at the same time as the samples and allow the operator to produce a standard curve of Optical Density versus F13B concentration. The concentration of in the samples is then determined by comparing the O.D. of the samples to the standard curve.

Principle of the Assay: The coated well immunoenzymatic assay for the quantitative measurement of F13B utilizes a multiclonal anti-F13B antibody and an F13B-HRP conjugate. The assay sample and buffer are incubated together with F13B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the F13B concentration since F13B from samples and F13B-HRP conjugate compete for the anti-F13B antibody binding site. Since the number of sites is limited, as more sites are occupied by F13B from the sample, fewer sites are left to bind F13B-HRP conjugate. Standards of known F13B concentrations are run concurrently with the samples being assayed and a standard curve is plotted relating the intensity of the color (Optical Density) to the concentration of F13B. The F13B concentration in each sample is interpolated from this standard curve.

Product Notes

The Human F13B f13b (Catalog #AAA86731) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA86731 ELISA Kit recognizes Human F13B. It is sometimes possible for the material contained within the vial of "Coagulation Factor XIII B Polypeptide, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
