Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific to Free-T3 has been pre-coated onto a microplate. Standards or samples are added to the appropriate microtiter plate wells with biotin-conjugated Free-T3 and incubated. A competitive inhibition reaction is launched between Free-T3 (Standards or samples) and Biotin-conjugated Free-T3 with the pre-coated antibody specific for Free-T3. The more amount of Free-T3 in samples, the less antibody bound by Biotin-conjugated Free-T3. Then Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of Free-T3 in the sample. The color development is stopped and the intensity of the color is measured.
