Principle of the Assay: The Mouse Gremlin 2 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse Gremlin 2 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Mouse Gremlin 2 coated on a 96-well plate. Standards and samples are pipetted into the wells and Gremlin 2 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse Gremlin 2 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Gremlin 2 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background: GREM2, also known as PRDC, is one of several BMP antagonists that regulate the functions of BMPs during embryonic development and tissue differentiation. BMP antagonists contain cystine knots and typically form homo- and heterodimers. The CAN (cerberus and dan) subfamily of BMP antagonists, to which this gene belongs, is characterized by a C-terminal cystine knot with an eight-membered ring. The antagonistic effect of the secreted glycosylated protein encoded by this gene is likely due to its direct binding to BMP proteins. The PRDC gene is mapped to chromosome 1q43.
