Biological Significance
IL6 mediates its biological effects through the type I IL6 receptor system that consists of two chains, IL6Ra and gp130. The IL6Ra chain is the binding component specific to IL6; while the gp130 only transmits signals of IL6 when bound to IL6Ra. The gp130 also can transmit signals from LIF, OSM, CNTF, IL11 and CT-1 in conjunction with other receptor subunits. The low-affinity binding site for IL6 is composed of IL6Ra alone. IL6Ra is expressed in a wide range of cells including T cells, fibroblasts and macrophages. Soluble IL6Ra which consists of only the extracellular domain of the IL6Ra chain, acts as an agonist of IL6 activity at low concentrations.
Bioactivity
The ED50 was determined by its ability to intensify the IL-6 induced growth inhibition of Mouse M1 cells is approximately 5.0 ng/ml, in the presence of 20 ng/ml of rhIL-6.
Endotoxin Levels
Endotoxin level is less than 0.1 ng per ug (1 EU/ug).
Expression System
HEK cells
Preparation and Storage
Store at 4 degree C until reconstitution. Following reconstitution aliquot and freeze at -20 degree C for long term storage. Avoid repeated freeze/thaw cycles.

It is sometimes possible for the material contained within the vial of "IL6R alpha, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
