Background: Calcifediol is a prehormone which is produced by hydroxylation of vitamin D3 (cholecalciferol) in the liver. Calcidiol is then converted in the kid neys (by the enzyme 25(OH)D D-1alpha-hydroxylase) into calcitriol (1,25-(OH)2D3), a secosteroid hormone that is the active form of vitamin D. It can also be converted into 24-hydroxycalcidiol in the kidneys via 24-hydroxylation. In medicine, blood concentration of calcidiol is considered the best indicator of vitamin D status. It is the most sensitive measure, though experts have called for improved standardization and reproducibility across different laboratories.

An antibody specific for 25 HVD3 has b een pre pre-coated onto a microplat microplate. Standards and samples are pipetted into the wells an d any 25 HVD3 present is bound by the immobi lized antibody. After removing any unbound substances substances, HRPHRP-ConjugateConjugated Human 25 HVD3 detection anti body is added to the we lls. Fo llowing a wash to r emove any unbound HRP reagent, a Ch romogen solution is adde d to the wells and color develops in proportion to the amount of 25 HVD3 bound in the i nitial s tep. The color development is st opped and the inten sity of th e color is measured.
