Intended Uses: The CD40L ELISA has been designed for the precise measurement of CD40L in human serum, body fluids, tissue homogenate or cell culture supernates. For research use only.
Principle of the Assay: This kit is based on a sandwich enzyme-linked immune-sorbent assay technology. An analyte-specific polyclonal antibody is pre-coated onto 96-well plates. The biotin conjugated second antibody is used as detection antibody. During the first incubation the standards and samples react with the analyte-specific pre-coated antibody. In a second incubation the biotin conjugated detection antibody completes the sandwich. After washing with wash buffer the TMB substrate is added to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The optical density of the yellow color is proportional to the analyte captured in plate. Read the O.D. absorbance at 450nm in a microtiterplate reader, and calculate the concentration of the analyte in the sample by taking into consideration the dilution factor of the sample!!Background/Introduction: CD40L, also called CD154, is a 261 amino acids protein is a member of the TNF superfamily of molecules. It is primarily expressed on activated T cells but is also found in a soluble form. It binds to CD40 on antigen-presenting cells (APC), which leads to many effects depending on the target cell type. In general, CD40L plays the role of a costimulatory molecule and induces activation in APC in association with T cell receptor stimulation by MHC molecules on the APC. In total CD40L has three binding partners: CD40, alpha5beta1 integrin and alphaIIbbeta3.
