Principle of the Assay: The AAA Biotech' Human CXCL10 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human CXCL10 in Cell Culture Supernatants, Serum, Plasma, Urine. This assay employs an antibody specific for Human CXCL10 coated on a 96-well plate. Standards and samples are pipetted into the wells and CXCL10 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human CXCL10 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CXCL10 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background/Introduction: Chemokine (C-X-C motif) ligand 10 (CXCL10) or IP-10 is a small cytokine belonging to the CXC chemokine family that is also known as 10 kDa interferon-gamma-induced protein (gamma-IP10 or IP-10). CXCL10 is secreted by several cell types in response to IFN-gamma. These cell types include monocytes, endothelial cells and fibroblasts. CXCL10 has been attributed to several roles, such as chemoattraction for monocytes/macrophages, T cells, NK cells, and dendritic cells, promotion of T cell adhesion to endothelial cells, antitumor activity, and inhibition of bone marrow colony formation and angiogenesis.It is a potent inhibitor of angiogenesis in vivo. The gene for CXCL10 is located on human chromosome 4 in a cluster among several other CXC chemokines. It is a RAS target gene and is overexpressed in the majority of colorectal cancers.This chemokine elicits its effects by binding to the cell surface chemokine receptor CXCR3.
