Background: The epidermal growth factor receptor(EGFR; ErbB-1; HER1 in humans) is the cell-surface receptor for members of the epidermal growth factor family(EGF-family) of extracellular protein ligands.1 It is a member of the ErbB family of receptors, a subfamily of four closely related receptor tyrosine kinases: EGFR(ErbB-1), HER2/c-neu(ErbB-2), Her 3(ErbB-3) and Her 4(ErbB-4). EGFR exists on the cell surface and is activated by binding of its specific ligands, including epidermal growth factor and transforming growth factor alpha(TGFalpha). EGFR and its ligands are cell signaling molecules involved in diverse cellular functions, including cell proliferation, differentiation, motility, and survival, and in tissue development. Mutations that lead to EGFR overexpression(known as upregulation) or overactivity have been associated with a number of cancers, including lung cancer and glioblastoma multiforme. In this latter case a more or less specific mutation of EGFR, called EGFRvIII is often observed.
Principle of the Assay: The Quick Picokine™ Human EGFR Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human EGFR in cell culture supernatants, serum, plasma (heparin, EDTA) and human milk. It uses our proprietary Quick ELISA technology. Quick ELISA technology employs capture antibodies conjugated to an affinity tag that is recognized by the polyclonal antibody used to coat our Quick ELISA plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. The kit includes Human EGFR protein as standards. To measure Human EGFR, add standards and samples to the wells, then add antibody cocktail. Wash the wells with PBS or TBS buffer, and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product is linearly proportional to Human EGFR in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human EGFR in the sample.
