Background: Basigin is a member of the immunoglobulin superfamily that is also known as EMMPRIN, short for extracellular matrix metalloproteinase inducer, and recently has been designated CD147(cluster of differentiation 147). It is a member of the immunoglobulin superfamily, with a structure related to the putative primordial form of the family. As members of the immunoglobulin superfamily play fundamental roles in intercellular recognition involved in various immunologic phenomena, differentiation, and development, basigin is thought also to play a role in intercellular recognition(Miyauchi et al., 1991; Kanekura et al., 1991).This protein is a determinant for the Ok blood group system. Basigin is a type I integral membrane receptor that has many ligands, including the cyclophilin(CyP) proteins Cyp-A and CyP-B and certain integrins. It is expressed by many cell types, including epithelial cells, endothelial cells and leukocytes.
Principle of the Assay: The Quick Picokine™ Human BSG Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human BSG in cell culture supernatants, serum, plasma (heparin, EDTA), saliva, urine and human milk. It uses our proprietary Quick ELISA technology. Quick ELISA technology employs capture antibodies conjugated to an affinity tag that is recognized by the polyclonal antibody used to coat our Quick ELISA plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. The kit includes Human BSG protein as standards. To measure Human BSG, add standards and samples to the wells, then add antibody cocktail. Wash the wells with PBS or TBS buffer, and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product is linearly proportional to Human BSG in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human BSG in the sample.
