Background: Eosinophil granule major basic protein encoded by EMBP/MBP gene is the predominant constituent of the crystalline core of the eosinophil granule. High levels of the proform of this protein are also present in placenta and pregnancy serum, where it exists as a complex with several other proteins including pregnancy-associated plasma protein A (PAPPA), angiotensinogen (AGT), and C3dg. This protein may be involved in antiparasitic defense mechanisms as a cytotoxin and helminthotoxin, and in immune hypersensitivity reactions.
Principle of the Assay: Human Eosinophil Granule Major Basic Protein (EMBP/MBP) ELISA Kit employs a two-site sandwich ELISA to quantitate EMBP/MBP in samples. An antibody specific for EMBP/MBP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any EMBP/MBP present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human EMBP/MBP detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of EMBP/MBP bound in the initial step.
