Principle of the Assay: The Picokine Human FGF9 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human FGF9 with a 96-well strip plate that is pre-coated with antibody specific for FGF9. The detection antibody is a biotinylated antibody specific for FGF9. The capture antibody is monoclonal antibody from mouse, the detection antibody is polyclonal antibody from goat. The kit contains recombinant Human FGF9 with immunogen: Expression system for standard: sf21; Immunogen sequence: M1-S208. The kit is analytically validated with ready to use reagents. To measure Human FGF9, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Human FGF9 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human FGF9 in the sample!!Background/Introduction: Fibroblast growth factor-9(FGF-9) is a steroid-regulated mitogen and survival factor for nerve and mesenchymal cells. The human FGF-9 cDNA cloned by using oligonucleotide probes encodes a polypeptide consisting of 208 amino acids. Sequence similarity to other members of the FGF family has been estimated to be around 30%. FGF-9 is an autocrine estromedin endometrial stromal growth factor that plays roles in cyclic proliferation of uterine endometrial stroma. FGF9 is produced and secreted by the prostatic stromal cells. It is a potent mitogen for both prostatic epithelial and stromal cells in culture. FGF9 is an abundant secreted growth factor that can act as both a paracrine mitogen for epithelial cells and an autocrine mitogen for stromal cells. Overexpression of this paracrine and autocrine growth factor may play an important role in the epithelial and stromal proliferation in benign prostatic hyperplasia.4 As a result of glycosylation, the molecular mass is 25-27KDa.
