Principle of the Assay: The interferons (IFN)s are a family of cytokines with potent antiviral, antiproliferative and immunomodulatory properties. The protein encoded by IFN-alpha gene is produced by macrophages and has antiviral activity.Human IFN-alpha ELISA Kit employs a double antibody sandwich method to quantitate Human IFN-alpha in samples. An antibody specific for Human IFN-alpha has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Human IFN-alpha present is bound by the immobilized antibody. After removing any unbound substances, a HRP-conjugated antibody specific for Human IFN-alpha is added to the wells. After washing, remove any unbound HRP-conjugated antibody reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measure the OD value with a microplate reader at 450 nm wavelength. The Human IFN-alpha concentration is proportional to the OD450 nm value.
