Specificity
The assay recognizes natural human IL-17A. To define specificity of this ELISA, several proteins were tested for cross reactivity. There was no cross reactivity observed for any protein tested (IL-1P, IL-6, IL-23, IFNy,IL-17B, IL-17D-::-::,IL-17E, IL-17F, IL-17A1F and murine IL-17A).

Intended Uses: The IL-12(P40) ELISA has been designed for the precise measurement of IL-12(P40) in human serum, plasma, body fluids, tissue homogenate or cell culture supernates. For research use only.
Principle of the Assay: This kit is based on a sandwich enzyme-linked immune-sorbent assay technology. An analyte-specific polyclonal antibody is pre-coated onto 96-well plates. The biotin conjugated second antibody is used as detection antibody. During the first incubation the standards and samples react with the analyte-specific pre-coated antibody. In a second incubation the biotin conjugated detection antibody completes the sandwich. After washing with wash buffer the TMB substrate is added to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The optical density of the yellow color is proportional to the analyte captured in plate. Read the O.D. absorbance at 450nm in a microtiterplate reader, and calculate the concentration of the analyte in the sample by taking into consideration the dilution factor of the sample!!Background/Introduction: Interleukin 12 (IL-12) is an interleukin that is naturally produced by dendritic cells, macrophages and human Blymphoblastoid cells (NC-37) in response to antigenic stimulation. IL-12 is composed of a bundle of four alpha helices. It is a heterodimeric cytokine encoded by two separate genes, IL-12A (p35) and IL-12B (p40). IL-12 plays an important role in the activities of natural killer cells and T lymphocytes. It mediates enhancement of the cytotoxic activity of NK cells and CD8+ cytotoxic T lymphocytes. IL-12 also has anti-angiogenic activity, which means it can block the formation of new blood vessels. IL12 p40 exists as a monomer and as a homodimer. The IL12B gene contains 8 exons, and maps to 5q31-q33.
