Background: Interleukin-8 (IL-8), also known as chemokine CXCL8, is a cytokine secreted by macrophages and epithelial cells. In recent years, people have found that IL-8 is almost involved in the whole reproductive process of mammals, such as ovulation, corpus luteum formation, endometrial proliferation, decidualization, embryo implantation and growth and development. Many clinical studies have shown that IL-8 is also related to some reproductive pathological processes. IL-8 works by binding to its specific receptor and triggering a series of biological events. IL-8 was initially characterized by its leukocyte chemotactic activity. Now it is known that IL-8 also has tumorigenic and angiogenic properties. This chemokine is secreted by a variety of cell types, including monocytes / macrophages, T cells, neutrophils, fibroblasts, endothelial cells and various tumor cell lines in response to inflammatory stimuli (IL1, TNF, LPS, etc.). At the same time, IL-8 / CXCL8 also has a wide range of effects on the above various types of cells, indicating that IL-8 / CXCL8 plays a crucial role in various pathological conditions such as chronic inflammation and cancer.
Principle of the Assay: IL-8 ELISA kit is applied to the solid phase sandwich Enzyme Linked Immunosorbent Assay. The microtiter plate has been pre-coated with capture antibody which specific to IL-8. Standards or samples are then added to the microtiter plate wells and IL-8 if present, will bind to the antibody pre-coated wells. Then a biotin-conjugated detection antibody specific for IL-8 is added to the wells to form a capture antibody-antigen-detection antibody sandwich complex. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells, and then a TMB substrate solution is added to the wells to incubate. TMB substrate solution turns blue after the oxidation of HRP, and finally turns to yellow immediately after adding the stop solution. Color of TMB substrate positively correlated with total IL-8 bound in the initial steps. The color is measured by spectrophotometrically with wavelength of 450nm. The concentration of IL-8 in samples is then determined by comparing the O.D. of the samples to the standard curve.
