Background: Leptospires ar e s piral-shaped, gram-negative obl igate aerobic s pirochetes w ith i nternal flagella. The genus is divided into two species, the pathogenic Leptospira interrogans and the free-living no npathogenic Leptospira bi flexa. Leptospira i nterrogans has abo ut 20 0 different serovars based on the variability of surface antigens. Leptospires affect mammals (wild and do mestic animals), r eptiles an d am phibians; t hey m ay be s hed i n t he ur ine lifelong. R ats an d o ther r odents ar e primary r eservoirs f or human i nfection w ith transmission being ef fected by ur ine-contaminated s oil or w ater, r at bi tes or i nfected animal tissues.
Principle of the Assay: The ELISA (Enzyme Li nked Immunosorbent A ssay) i s a n i mmunoassay, w hich i s particularly s uited t o t he d etermination of antibodies i n v arious k inds of s amples. T he reaction is based on the specific interaction of antibodies with their corresponding antigen. The test strips of the microtiter plate are coated with specific antigens of the pathogen of interest. I f ant ibodies i n t he s ample are present, they bi nd t o t he f ixed ant igen. A secondary antibody, which has been conjugated with t he enzyme alkaline phosphatase, detects a nd binds t o t he i mmune c omplex. T he c olourless s ubstrate p - nitrophenylphosphate i s t hen c onverted i nto t he c oloured pr oduct p -nitrophenol. T he signal intensity of this reaction product is proportional to the concentration of the analyte in the sample and is measured photometrically.
