Principle of the Assay: Human Matrix Metalloproteinase-26 (MMP26) ELISA Kit employs a two-site sandwich ELISA to quantitate MMP26 in samples. An antibody specific for MMP26 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP26 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human MMP26 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of MMP26 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background/Introduction: Matrix metalloproteinase-26 also known as matrilysin-2 and endometase is an enzyme that in humans is encoded by the MMP26 gene. MMP-26 is the smallest member of the matrix metalloproteinase (MMP) family of zinc-dependent endopeptidases. Synthesized as a zymogen, the nascent form is composed of three domains: "pre" domain, N-terminal signal sequence, which directs the protein into the endoplasmic reticulum; MMP-26 has been found strictly expressed in normal tissues of the placenta and moderately expressed in the uterus.
