Background: Matrix metalloproteinases are a family of zinc and calcium dependent endopeptidases with the combined ability to degrade all the components of the extracellular matrix. MMP-10 (stromelysin 2) degrades a broad range of substrates including gelatin, collagen types III, IV and V, fibronectin, aggrecan, and pig cartilage proteoglycan. MMP-10 can activate other MMPs such as MMP-1 and MMP-8. MMP-10 is expressed in keratinocytes, T cells, menstrual endometrium, and a few tumor samples. Structurally, MMP-10 may be divided into four distinct domains: a pro-domain which is cleaved upon activation, a catalytic domain containing the zinc binding site; a short linker region, and a carboxyl terminal hemopexin-like domain.
Principle of the Assay: The Human MMP10 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human MMP10 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Human MMP10 coated on a 96-well plate. Standards and samples are pipetted into the wells and MMP10 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human MMP10 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MMP10 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
