Principle of the Assay: The Human RSPO1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human RSPO1 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Human RSPO1 coated on a 96-well plate. Standards and samples are pipetted into the wells and RSPO1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human RSPO1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of RSPO1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background/Introduction: R-spondin-1 is a secreted protein that in humans is encoded by the Rspo1 gene. It is mapped to chromosome 1p34. This gene encodes a secreted activator protein with two cysteine-rich, furin-like domains and one thrombospondin type 1 domain. The encoded protein is a ligand for leucine-rich repeat-containing G-protein coupled receptors (LGR proteins) and positively regulates the Wnt signaling pathway. In mice, the protein induces the rapid onset of crypt cell proliferation and increases intestinal epithelial healing, providing a protective effect against chemotherapy- induced adverse effects. Alternative splicing results in multiple transcript variants.
