Background: Tapasin, or TAPBP (MIM 601962), is a member of the variable-constant Ig superfamily that links major histocompatibility complex (MHC) class I molecules to the transporter associated with antigen processing (TAP; see MIM 170260) in the endoplasmic reticulum(ER). The TAPBP gene is located near the MHC complex on chromosome 6p21.3. TAPBPL is a member of the Ig superfamily that is localized on chromosome 12p13.3, aregion somewhat paralogous to the MHC.
Principle of the Assay: Human Tapasin-related protein (TAPBPL) ELISA Kit employs a two-site sandwich ELISAto quantitate TAPBPL in samples. An antibody specific for TAPBPL has been pre-coatedonto a microplate. Standards and samples are pipetted into the wells and any TAPBPLpresent is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human TAPBPL detection antibody is added to the wells. Following awash to remove any unbound HRP reagent, a Chromogen solution is added to the wellsand color develops in proportion to the amount of TAPBPL bound in the initial step. Thecolor development is stopped and the intensity of the color is measured.
