Background: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of Thrombopoietin/Tpo in human serum, plasma, cell culture supernatants and other biological fluids.
Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. An antibody specific for human Thrombopoietin/Tpo has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Thrombopoietin/Tpo present is bound by the immobilized antibody. After washing away any unbound substances, a biotin-conjugated detection antibody specific for human Thrombopoietin/Tpo is added to the wells. After incubating, an enzyme-linked streptavidin is added and binds to biotin. Following a wash to remove any unbound antibody-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Thrombopoietin/Tpo bound in the initial step. The color development is stopped, and the absorbance is measured.
