Background: Bovine Immunoglobmlin G (IgG) is the major component of immunoglobulin in bovine serum with a molecular weight of about 160 kDa, 40? 50% of which are distributed in serum, and the rest are distributed in tissues. IgG is divided into 4 subtypes: IgG1, IgG2, IgG3 and IgG4, and its content in serum is 17-27mg/mL. Bovine immunoglobulin G (Bovine IgG) is widely used in basic biochemistry, research and teaching, biomedical production fields. The products produced by biotechnological processes, such as the materials used in cells or tissue culture media, all may lead to residual contamination of the finished products. Bovine IgG is often used as a medium to culture mammalian cells in recombinant pharmaceutical production because of it's large amout in fetal bovine serum. During the production process of recombinant drugs, some of the bovine IgG are removed by process optimization and purification, but some of them may still exist in the some of the recombinant drugs and other finished products. However when these products are used to treat for human or animals, bovine IgG as a sensitizers may cause an immune response, influence human health or even threaten lives at worst. Therefore, the detection of residual bovine IgG is an important process in the quality control of biomedical products.

Principle of the Assay: This ELISA kit is applied to the double antibody sandwich Enzyme Linked Immunosorbent Assay to detect the concentration of bovine IgG in samples. The microtiter plate has been pre-coated with rabbit anti-bovine antibody, standards or samples are then added to the microtiter plate wells and IgG if present, will bind to the antibody pre-coated wells under specific conditions. After washing, then a coupled biotin-conjugated detection rabbit anti-bovine is added to the wells, under 37? incubation, to form a precoated antibody-bovine IgG-detection antibody sandwich complex. After washing, coupled avidin conjugated Horseradish Peroxidase (HRP) is added to the wells, resulting in a cascade amplification effect. Then a TMB substrate solution is added to the wells to incubate. TMB substrate solution turns blue after the oxidation of HRP, and finally turns to yellow immediately after adding the stop solution. Color of TMB substrate positively correlated with total IgG bound in the initial steps. The color is measured by spectrophotometrically with wavelength of 450nm. The concentration of bovine IgG in samples is then determined by comparing the O.D. of the samples to the standard curve.
