Background: Mouse immunoglobulin G (IgG) is the main component of immunoglobulin in serum, it is mainly created and secreted by plasma cells in spleen and lymph nodes and exists in the form of monomer. IgG comprises approximately 75% of serum immunoglobulin. Its normal value is in mg/ml in body fluids with 40%-50% distributed in serum and the rest distributed in tissues. Immunoglobulin in the immune serum of human and animals has very dissimilar concentration and structure, there is considerable discrepancy with their composition, structure, size, electric charge, and biological activity, etc.The immunoglobulin comprises about 20-25% of whole serum albumin. To date, 5 types of immunoglobulin have been isolated and purified in human, mouse and other animals' serum-IgG, IgM, IgA, IgD and IgE. According to the antigenic specificity of gamma chain in IgG molecule, mouse IgG is divided into 4 subtypes: IgG1, IgG2a, IgG2b and IgG3. Mouse IgG can activate complement through classical pathway, its ability to bind complement is IgG2b?IgG2a?IgG3, while IgG1 has no ability to bind complement.

Principle of the Assay: This ELISA kit is applied to the double antibody sandwich Enzyme Linked Immunosorbent Assay to detect the concentration of mouse IgG in samples. The microtiter plate has been pre-coated with goat anti-mouse antibody, standards or samples are then added to the microtiter plate wells and IgG if present, will bind to the antibody pre-coated wells under specific conditions. After washing, then a coupled biotin-conjugated detection goat anti-mouse antibody is added to the wells, under 37? incubation, to form a precoated antibody-mouse IgG-detection antibody sandwich complex. After washing, coupled avidin conjugated Horseradish Peroxidase (HRP) is added to the wells, resulting in a cascade amplification effect. Then a TMB substrate solution is added to the wells to incubate. TMB substrate solution turns blue after the oxidation of HRP, and finally turns to yellow immediately after adding the stop solution. Color of TMB substrate positively correlated with total IgG bound in the initial steps. The color is measured by spectrophotometrically with wavelength of 450nm. The concentration of mouse IgG in samples is then determined by comparing the O.D. of the samples to the standard curve.
