Principle of the Assay: Interleukin 12 (IL-12), also known as natural killer cell stimulatory factor (NKSF) or cytotoxic lymphocyte maturation factor (CLMF),is a heterodimeric pleiotropic cytokine made up of a 40 kDa (p40) subunit and a 35 kDa (p35) subunit.Mouse IL-12 p70ELISA Kit employs a double antibody sandwich method to quantitate Mouse IL-12 p70 in samples. An antibody specific for MouseIL-12 p70 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Mouse IL-12 p70present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific forMouse IL-12 p70 is added to the wells. After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Following awash to remove any unbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis ofHRP, and turns yellow after adding stop solution. Measure the OD value with a microplate reader at 450nm wavelength. The IL-12p70 concentration is proportional to the OD450 nm value.
