Background: Insulin is a peptide hormone produced by INS encoded human pancreatic islet beta cells, single-chain insulin propeptide consists of a 30 amino acid B chain (aa 2554), a C peptide segment (aa 5589), and a 21 amino acid A chain (aa 90110). After removal of C-peptide by protease hydrolysis, it becomes an active form, that is, a disulfide heterodimer composed of A chain and B chain. It is considered the main anabolic hormone of the body.It stimulates the glucose absorption from the blood into the liver, fat and skeletal muscle cells, so as to adjust the metabolism of carbohydrates, fat and protein. The high concentrated insulin in the blood strongly inhibits the production and secretion of glucose from the liver. Circulating insulin also affects the synthesis of various tissue proteins. Therefore, it is an anabolic metabolism hormone, which can promote the conversion of small molecules in the blood to larger big molecules within cells. The low concentrated insulin has the opposite effect by promoting extensive catabolism, especially for body fat storage. Diseases known to be associated with insulin include hyperinsulinemia and type 10 maturity-onset diabetes of youth. Multiple mutant alleles with phenotypic effects have been identified, including insulin-dependent diabetes, permanent neonatal diabetes, type 10 maturity-onset diabetes of youth, and hyperinsulinemia.

Principle of the Assay: This ELISA kit is applied to the solid phase sandwich Enzyme Linked Immunosorbent Assay. The microtiter plate has been pre-coated with capture antibody. The antigen to be measured and the biotin-conjugated detection antibody are then added to the microtiter plate wells sequentially to form a capture antibody-antigen-detection antibody complex. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells, and then a TMB substrate solution is added to the wells to incubate. TMB substrate solution turns blue after the oxidation of HRP, and finally turns to yellow immediately after adding the stop solution. Color of TMB substrate positively correlated with total insulin bound in the initial steps. The color is measured by spectrophotometrically with wavelength of 450nm. The concentration of insulin in samples is then determined by comparing the O.D. of the samples to the standard curve.
