Background: Insulin (INS) is a protein hormone produced by the beta cells of the pancreas. It plays a crucial role in regulating glucose metabolism. INS lowers blood glucose levels by promoting the uptake and utilization of glucose by cells, and by promoting the storage of glucose in the liver and muscles. It also promotes the synthesis of proteins and fats, and inhibits the breakdown of proteins and fats, to maintain a balance of nutrients within the body. Insufficient levels of INS can lead to diabetes. Type 1 diabetes is caused by an absolute deficiency of insulin, usually due to the immune system attacking and destroying beta cells in the pancreas. Type 2 diabetes, on the other hand, is caused by insufficient insulin or insulin resistance, and is often related to obesity, lifestyle, and genetic factors.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti INS antibody was pre-coated onto the 96-well plate. The biotin conjugated anti INS antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with INS conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of INS in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
