Introduction: Matrix metallopeptidase 1, also known as MMP-1, is a member of the matrix metalloproteinase (MMP) family. MMP family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. MMP-1 gene is part of a cluster of MMP genes which localize to chromosome 11q22.3. The enzyme is expressed by a wide variety of cell types. MMP-1 is the only enzyme able to initiate breakdown of the interstitial collagens, collagen type 1, collagen type 2, and collagen type 3. MMP-1 plays an important role in diverse physiologic processes such as development, tissue morphogenesis, and wound repair. MMP-1 is synthesized as an inactive proform that requires proteolytic cleavage of N-terminal residues for activation. MMP-1 plays an important role in the remodeling of collagenous extracellular matrix in various physiologic and pathologic situations. MMP-1 also cleaves a number of non-matrix substrates and cell surface molecules, suggesting a role in the regulation of cellular behaviour in addition to its role in the turnover of collagen fibrils.

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific to MMP-1 has been pre-coated onto a microplate. Standards or samples are added to the appropriate microtiter plate wells with biotin-conjugated MMP-1 and incubated. A competitive inhibition reaction is launched between MMP-1 (Standards or samples) and Biotin-conjugated MMP-1 with the pre-coated antibody specific for MMP-1. The more amount of MMP-1 in samples, the less antibody bound by Biotin-conjugated MMP-1. Then Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of MMP-1 in the sample. The color development is stopped and the intensity of the color is measured.
