Product Notes

The AHR ahr (Catalog #AAA24424) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The AHR (Aryl Hydrocarbon Receptor, Ah Receptor, AhR, Class E Basic Helix-loop-helix Protein 76, bHLHe76, BHLHE76) (APC) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's AHR can be used in a range of immunoassay formats including, but not limited to, FLISA, IF (Immunofluorescence), IP (Immunoprecipitation), WB (Western Blot). Applications are based on unconjugated antibody. Researchers should empirically determine the suitability of the AHR ahr for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "AHR, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.

Frequently Asked Questions

What cell types or tissues express human AHR that this antibody can detect?

The Aryl Hydrocarbon Receptor (AHR) is widely expressed across tissues, including intestinal epithelium, immune cells (T cells, dendritic cells, macrophages), liver, lung, skin, and nervous tissue. This antibody detects endogenous human AHR protein across diverse cell types and tissues, enabling comprehensive assessment of AHR expression patterns.

Can this anti-AHR antibody be used for Western blot, flow cytometry, or immunofluorescence?

Yes, this anti-AHR antibody is validated for multiple applications: Western blotting (quantifying AHR protein levels ~110 kDa), flow cytometry (identifying AHR+ immune cell populations), and immunofluorescence (assessing subcellular AHR localization). These complementary techniques provide a comprehensive characterization of AHR expression and activation status.

How does AHR activation affect gene expression or immune responses?

AHR is a ligand-activated transcription factor that, upon activation, translocates to the nucleus, forms heterodimers with ARNT, and drives expression of xenobiotic metabolism genes (CYP1A1, CYP1B1) plus immunomodulatory genes (IL-22, IL-17). AHR activation profoundly shapes Th17 cell differentiation, barrier immunity, and tumor-associated immune suppression.

Does this antibody detect ligand-bound versus unbound AHR forms?

This antibody typically detects total AHR protein regardless of ligand-binding status. However, AHR conformation changes upon activation (ligand binding and nuclear translocation) may affect antibody accessibility. Some applications (immunofluorescence showing nuclear vs. cytoplasmic distribution) indirectly assess AHR activation state based on subcellular localization patterns.

Is AHR expression upregulated by environmental toxins or xenobiotics?

AHR expression itself is not dramatically upregulated by xenobiotics, but AHR target gene expression is strongly activated by ligand binding. Classical xenobiotics (TCDD/dioxin, PAHs) potently activate AHR signaling. Importantly, endogenous AHR ligands (dietary components, microbial metabolites like kynurenine) and emerging environmental chemicals also activate AHR-dependent responses.

Can this antibody be used to study AHR signaling in cancer or immune regulation?

Yes, this antibody enables comprehensive AHR signaling studies in both contexts. In cancer, AHR shows context-dependent roles (tumor-suppressive in lung cancer, tumor-promoting in other contexts). In immunity, AHR profoundly regulates T cell differentiation, barrier immunity, and tumor immune escape, making this antibody invaluable for dissecting AHR's complex immunological functions.
