DnaK possesses an autophosphorylation activity and a weak 5'-nucleotidase activity, cleaving the 5'-phosphate groups from both ribose- and deoxyribose nucleotides. Genetic and biochemical studies have demonstrated protein-protein interactions between the DnaK protein and the lambdaO and lambdaP DNA replicating proteins. DnaK has also been shown to interact with DnaJ and GrpE heat shock proteins. In addition, DnaK has been demonstrated to preferentially bind to the unfolded form of bovine pancreatic trypsin inhibitor as opposed to the native protein. The role of DnaK in ATP-dependent protein-protein interactions has been extended to normal E. coli physiology. Like eukaryotic Hsp70 homologs, it is thought to participate in the assembly/disassembly of protein complexes, essentially as first proposed by Hightower and Pelham.
