FCM/FACS (Flow Cytometry)

(Flow cytometric analysis of 2% paraformaldehyde-fixed Jurkat (Human T cell leukemia cells from peripheral blood) cells labeling G3BP1 with M25069 at 1/200 dilution (red) compared with a mouse monoclonal IgG isotype control (blue) and an unlabelled control (cells without incubation with primary antibody, green). Goat anti-mouse IgG (FITC) at 1/300 dilution was used as the secondary antibody.)

IP (Immunoprecipitation)

(G3BP1 was immunoprecipitated from 1mg of Jurkat cells membrane fraction, blotted with M25069 of 10 ug. Western blot was performed from the immunopre-cipitate using M25069 at 1/1000 dilution. Anti-Mouse-IgG(HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/10000 dilution. Lane 1: Jurkat cells membrane fraction. This gene encodes one of the DNA-unwinding enzymes which prefers partially unwound 3'-tailed substrates and can also unwind partial RNA, DNA and RNA, RNA duplexes in an ATP-dependent fashion. This enzyme is a member of the heterogeneous nuclear RNA-binding proteins and is also an element of the Ras signal transduction pathway. It binds specifically to the Ras-GTPase-activating protein by associating with its SH3 domain. Several alternatively spliced transcript variants of this gene have been described, but the full-length nature of some of these variants has not been determined.
