Specificity
A sensitive antibody to Leishmania amastigote-specific A2 protein. This sequence can also be used as tag if cloned with the protein of interest. This sequence is particular to the parasite and therefore the antibody has no crossreactivity with mammalian proteins. It is specific to A2 protein from Leishamania donovani and in a highly probable manner to other species which express A2. It won't be detected in species where the A2 is non-expressed pseudogenes such as Leishmania major.

The anti-A2 antibody recognizes only the amastigote form of the A2 protein. The anti-A2 antibody recognizes the sequence QSVGPLSVGP from the parasite, Leishmania donovani.

Preparation and Storage
Lyophilized antibodies can be kept at 4ºC for up to 3 months and should be kept at -20ºC for long-term storage (2 years). To avoid freeze-thaw cycles, reconstituted antibodies should be aliquoted before freezing for long-term (1 year) storage (-80ºC) or kept at 4ºC for short-term usage (2 months). For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made with the assay buffer. After the maximum long-term storage period (2 years lyophilized or 1 year reconstituted) antibodies should be tested in your assay with a standard sample to verify if you have noticed any decrease in their efficacy. To limit antibody loss or degradation, BSA (final concentration 1%) and sodium azide (final concentration 0.02%) can be added to the suggested first dilution. It is important to first verify if those preservatives are compatible with your assay.

Leishmania donovani is a protozoa parasite responsible for human visceral leishmaniasis. During their life cycle the parasites exist in 2 forms: as flagellated promastigote within the sandfly vector and as non-flagellated amastigote in the macrophage phagolysosomal compartment of the mammalian host. A2 protein is an amastigote-specific virulence factor that is required for Leishmania donovani parasite survival in a mammalian host.
