Specificity
Tumor Necrosis Factor Alpha (TNF alpha) is a protein secreted by lipopolysaccharide-stimulated macrophages, and causes tumor necrosis when injected into tumor bearing mice. TNF alpha is believed to mediate pathogenic shock and tissue injury associated with endotoxemia. TNF alpha exists as a multimer of two, three, or five non-covalently linked units, but shows a single 17kDa band following SDS PAGE under non-reducing conditions. TNF alpha is closely related to the 25kDa protein Tumor Necrosis Factor beta (lymphotoxin), sharing the same receptors and cellular actions. TNF alpha causes cytolysis of certain transformed cells, being synergistic with interferon gamma in its cytotoxicity. Although it has little effect on many cultured normal human cells, TNF alpha appears to be directly toxic to vascular endothelial cells. Other actions of TNF alpha include stimulating growth of human fibroblasts and other cell lines, activating polymorphonuclear neutrophils and osteoclasts, and induction of interleukin 1, prostaglandin E2 and collagenase production.

Product Notes

The TNF tnf (Catalog #AAA13806) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The TNF-alpha (Tumor Necrosis Factor alpha) Mouse Monoclonal Antibody reacts with Human, Mouse, Rat, Rabbit, Cat, Dog, Zebrafish and may cross-react with other species as described in the data sheet. AAA Biotech's TNF-alpha (Tumor Necrosis Factor alpha) can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC), Immunofluorescence (IF), Immunohistochemistry (IHC) Formalin/Paraffin. Flow Cytometry (0.5-1ug/million cells) Immunofluorescence (1-2ug/ml) Immunohistochemistry (Formalin-paraffin) (2-4ug/ml for 30 min at RT) (Staining of formalin-fixed tissues requires boiling tissue sections in 10mM Tris Buffer with 1mM EDTA, pH 9.0, for 10-20 min followed by cooling at RT for 20 minutes) Optimal dilution for a specific application should be determined. Researchers should empirically determine the suitability of the TNF tnf for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TNF-alpha (Tumor Necrosis Factor alpha), Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
