ICC (Immunocytochemistry)

(Immunocytochemistry analysis of MCF7 cells labeling Topoisomerase I with Rabbit anti- Topoisomerase I antibody at 1/1,000 dilution. Cells were fixed in 4% paraformaldehyde for 20 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 5 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Rabbit anti-Topoisomerase I antibody at 1/1,000 dilution in 1% BSA in PBST overnight at 4 degree C. Goat Anti-Rabbit IgG H&L (iFluor 488) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI. Beta tubulin (red) was stained at 1/100 dilution overnight at +4 degree C. Goat Anti-Mouse IgG H&L (iFluor 594) was used as the secondary antibody at 1/1,000 dilution.)

IHC (Immunohiostchemistry)

(Immunohistochemical analysis of paraffin-embedded human large intestine tissue with Rabbit anti-Topoisomerase I antibody at 1/1,000 dilution. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/1,000 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.)

Related Product Information for anti-TOP1 antibody
This gene encodes a DNA topoisomerase, an enzyme that controls and alters the topologic states of DNA during transcription. This enzyme catalyzes the transient breaking and rejoining of a single strand of DNA which allows the strands to pass through one another, thus altering the topology of DNA. This gene is localized to chromosome 20 and has pseudogenes which reside on chromosomes 1 and 22. [provided by RefSeq, Jul 2008]

Product Notes

The TOP1 top1 (Catalog #AAA268393) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Topoisomerase I reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Topoisomerase I can be used in a range of immunoassay formats including, but not limited to, FCM/FACS (Flow Cytometry), IHC (Immunohistochemistry), IF (Immunofluorescence), ICC (Immunocytochemistry), WB (Western Blot). Researchers should empirically determine the suitability of the TOP1 top1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Topoisomerase I, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
