Background: C3 is the complement component with the highest content in serum, which is mainly synthesized by macrophages and liver. Under the action of C3 invertase, C3 is cleaved into two fragments, C3a and C3b, which play an important role in both the classical and bypass activation pathways of complement. Increased C3 is commonly seen in some acute inflammation or early infectious diseases, such as acute rheumatic fever, myocarditis, myocardial infarction, arthritis and so on. C3 reduction is commonly seen in decreased complement synthesis, such as chronic active hepatitis, cirrhosis, liver necrosis, etc. Excessive depletion or loss of complement, such as active lupus erythematosus, early and late acute glomerulonephritis, basement membrane proliferative glomerulonephritis, cryoglobulinemia, severe rheumatoid arthritis, extensive burns, etc.

Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti C3 antibody was precoated onto the 96-well plate. The biotin conjugated anti C3 antibody was used as the detection antibody. The standards and pilot samples were added to the wells subsequently. After incubation, unbound conjugates were removed by wash buffer. Then, biotinylated detection antibody was added to bind with C3 conjugated on coated antibody. After washing off unbound conjugates, HRP-Streptavidin was added. After a third washing, TMB substrates were added to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that turned yellow after adding a stop solution. Read the O.D. absorbance at 450nm in a microplate reader. The concentration of C3 in the sample was calculated by drawing a standard curve. The concentration of the target substance is proportional to the OD450 value.
