Principle of the Assay: Granulocyte-macrophage colony-stimulating factor (GM-CSF), also known as colony stimulating factor 2 (CSF2), is a monomericglycoprotein secreted by macrophages, T cells, mast cells, NK cells, endothelial cells and fibroblasts that functions as a cytokine.. Mouse GM-CSF ELISA Kit employs a double antibody sandwich method to quantitate Mouse GM-CSF in samples. Anantibody specific for Mouse GM-CSF has been pre-coated onto a microplate. Standards and samples are pipetted into the wellsand any Mouse GM-CSF present is bound by the immobilized antibody. After removing any unbound substances, abiotin-conjugated antibody specific for Mouse GM-CSF is added to the wells. After washing, proprietary Streptavidin-HRPconjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent, adding HRP Substrate(TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measure the OD value with amicroplate reader at 450 nm wavelength. The GM-CSF concentration is proportional to the OD450 nm value.
