Principle of the Assay: Interleukin 17 (IL-17; also known as IL-17A) is a 30-35 kDa variably glycosylated homodimeric protein that belongs to a uniquefamily of cysteine-knot related proteins. Its sequence was originally isolated from an activated rodent hybridoma and termedCTLA-8.Mouse IL-17 ELISA Kit employs a double antibody sandwich method to quantitate Mouse IL-17 in samples. Anantibody specific for Mouse IL-17 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells andany Mouse IL-17 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugatedantibody specific for Mouse IL-17 is added to the wells. After washing, proprietary Streptavidin-HRP conjugates is added to thewells. Following a wash to remove any unbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blueunder the catalysis of HRP, and turns yellow after adding stop solution. Measure the OD value with a microplate reader at 450nmwavelength. The IL-17 concentration is proportional to the OD450 nm value.
